About Liquid-State NMR Spectroscopy
The vast majority of NMR samples are studied in the liquid-state where solid samples are dissolved in the appropriate deuterated solvent. In order to study a sample by liquid-state NMR spectroscopy:
- Enough sample must be provided to make a 0.6 mL solution of at least 1mM. Usually, more sample is required and the solution concentration required will depend on the desired NMR nucleus of interest and experiment. In most cases, solutions of at least 10mM are preferred. If your sample is not appreciably soluble in any known solvent, you may consider using solid-state NMR spectroscopy instead.
- The sample should be diamagnetic. Paramagnetic samples can be studied by NMR, but the NMR is much more complicated and may not give rise to any useable data.
Services Provided
Depending on your experience using NMR spectroscopy as a means of structural characterization, the NMR facility can perform a variety of functions. For experienced users, staff generally just acquire the NMR data and provide the raw NMR data. For novice users, the NMR facility can prepare the NMR sample, acquire the NMR data, process, and completely analyze the NMR spectra. For all external service work, a report summarizing the NMR experiments performed and the experimental conditions utilized are included. Experiments are typically completed within 48 hours of sample receipt. If the complete analysis is required, at least 1 week is necessary.
The NMR facility provides a base service for all samples, which includes:
- NMR tube
- NMR solvent (deuterated solvent)
- preparation of solution for NMR
- acquisition of a 1D 1H NMR spectrum at 25 oC
- e-mailing of raw NMR data (FID)
- service summary report outlining the NMR experiments performed, NMR conditions and charges (external samples only)
- sample disposal
For an additional charge above that of the base service, the NMR facility can perform the following NMR experiments:
- Standard direct-detect 1D NMR experiments on 13C, 19F, and 31P.
- Odd-nuclei direct-detect 1D NMR experiments on nuclear isotopes such as 2H, 11B, 77Se. Many other nuclear isotopes can be studied as well. Please enquire.
- Indirect-detect heteronuclear 2D NMR experiments on nuclear isotopes such as 15N, 29Si. Many other nuclear isotopes can be studied as well. Please enquire.
- Homonuclear 2D 1H-1H NMR experiments such as COSY, TOCSY, NOESY/ROESY.
- Any of the above experiments can also be performed above or below room temperature.
For an additional charge above that of the base service, the NMR facility will perform or help with the following tasks:
- Processing of the NMR data into an NMR spectrum.
- Analysis of the NMR spectra, including integration, peak picking, and peak assignments where applicable.
- Interpretation of 1D and 2D NMR spectra.

Sample Submission Procedures
